Abstract

Low temperature stress is one of the major environmental factors limiting the development of citrus cultivation and production. In this study, the effect of low temperature stress on the biochemical and physiological indices of Japanese tangerine and Star Ruby grapefruit grafted onto sour orange rootstock (two-year-old seedlings) was investigated. The experiment was conducted as a completely randomized design with cold temperature treatments (-8, -4, and 4 °C). Comparison of treatment means showed that the highest leaf ion leakage and the lowest chlorophyll a, carotenoid, and total chlorophyll pigment contents were obtained at -8 °C in grapefruit. Relative water content decreased with decreasing temperature, reaching its lowest level at -8 °C. Furthermore, the highest levels of lipid peroxidation (1.55 µg·g⁻¹ FW MDA) and total flavonoids (30.42 mg·g⁻¹ FW quercetin equivalents) were recorded at -4 °C in grapefruit. The highest activities of catalase (36.36 µmol·g⁻¹ FW·min⁻¹), ascorbate peroxidase (59.3 enzyme units·mg⁻¹ FW), and superoxide dismutase (161.3 µmol·g⁻¹ FW) enzymes were observed at -8 °C, in tangerine, grapefruit and tangerine respectively. Japanese mandarin seedlings grafted onto sour orange rootstock showed no signs of damage at -8°C during the cold acclimation phase. The seedlings resumed normal growth after the treatment period and recovery from freezing stress. In contrast, 'Ruby Star' grapefruit seedlings exhibited signs of damage such as leaf curling in the majority of their leaves at -8°C.
