Abstract

To investigate the effects of spermidine on selected morphological, physiological, and biochemical traits of two grapevine cultivars (Siah Sardasht and White Seedless) under salinity stress, a six-month greenhouse experiment was conducted using a factorial arrangement in a completely randomized design with four replicates. The experiment included three factors: grapevine cultivar (two levels: Siah Sardasht and White Seedless), salinity (four levels: 0 (control), 20, 40, and 80 mM sodium chloride (NaCl) added to the nutrient solution), and foliar spermidine application (four levels: 0 (control), 0.25, 0.5, and 1 mM). At 80 mM salinity, plant height in White Seedless decreased by 26.7% compared with the control. Under salinity levels of 40 and 80 mM, application of 1 mM spermidine resulted in reductions in leaf relative water content of 4.4% and 11.64%, respectively, compared with the control. Soluble sugar content at 80 mM salinity increased 2.6-fold in White Seedless and 1.6-fold in Siah Sardasht compared with the respective controls. In White Seedless, ascorbate peroxidase activity increased 2.36-fold and 2.92-fold under 40 and 80 mM salinity, respectively, relative to the control. The findings indicated that White Seedless was more tolerant to salinity than Siah Sardasht for several measured traits. Foliar application of spermidine may alleviate some adverse effects of salinity stress in both grapevine cultivars.
