Abstract

Pomegranate is one of the most important horticultural crops, and Iran is a major producer worldwide. Salinity, meanwhile, is one of the most significant environmental constraints in arid and semi-arid regions. The use of biostimulants to mitigate the adverse effects of environmental stresses, including salinity, has recently increased. This study investigated the effects of pretreatment with different concentrations of chitosan, melatonin, and hydroxypropyl methylcellulose (HPMC) on two-year-old pomegranate seedlings of the Malas-e Saveh cultivar under salinity stress. The experiment was conducted in a research greenhouse at the University of Maragheh using a factorial arrangement in a completely randomized design with three replicates. The first factor was salinity at two levels (0 and 100 mM NaCl). The second factor comprised pretreatments with melatonin at 50 mg L⁻¹, chitosan, HPMC, a combination of melatonin coated with chitosan and HPMC (each at 100 mg L⁻¹), and distilled water as the control. Salinity stress reduced leaf number, photosynthetic pigment content, and membrane stability, while increasing proline content, superoxide dismutase activity, and malondialdehyde content. However, combined pretreatment with HPMC, melatonin, and chitosan increased leaf number, photosynthetic pigment content, membrane stability, and antioxidant enzyme activity, while reducing malondialdehyde content. Melatonin pretreatment alone also positively affected plant growth and reduced hydrogen peroxide content. Based on these findings, pretreatment with HPMC, melatonin, and chitosan (particularly melatonin coated with chitosan and HPMC) is recommended to mitigate the adverse effects of salinity stress.
