Abstract

This study evaluated the micropropagation potential of four promising apple rootstocks (AR11, AR10, AR8, and AR4) and the Azayesh Isfahan cultivar. Explants obtained from the selected rootstocks were established under in vitro conditions, and two experiments were conducted to identify the most suitable culture media and plant growth regulator treatments. In the first experiment, after explant establishment on MS medium at half-strength concentration (½ MS), the effects of four concentrations of 6-benzylaminopurine (BAP; 0.5, 1.0, 1.5, and 2.0 mg L⁻¹) on shoot proliferation were evaluated. In the second experiment, root formation was assessed using indole-3-butyric acid (IBA) and 1-naphthaleneacetic acid (NAA), each at concentrations of 1 and 2 mg L⁻¹. The highest number of shoots was obtained in hybrid genotype AR10 treated with 0.5 mg L⁻¹ BAP, with a mean of 9.26 shoots. The greatest shoot length was recorded in hybrid genotype AR8 under the same BAP treatment, averaging 2.80 cm. The highest fresh shoot weight (0.68 g) was observed in Azayesh Isfahan treated with 1.5 mg L⁻¹ BAP. The greatest root number and root length were obtained in hybrid genotype AR11 treated with 2 mg L⁻¹ NAA, with means of 10.33 roots and 9.33 cm, respectively. Among the genotypes evaluated, hybrid rootstocks AR11 and AR10 showed better shoot proliferation and rooting than the other genotypes when treated with 0.5 mg L⁻¹ BAP and 2 mg L⁻¹ NAA, respectively.
