Abstract

Walnut is considered one of the economically important crops with high nutritional value. Asexual propagation methods in walnut have failed to achieve large-scale production of walnut plantlets because of the presence of phenolic compounds that inhibit cell division. Walnut tissues are rich in phenolic compounds, which become dark after being excised from the mother plant as explants and, upon diffusion into the culture medium, severely affect explant survival and establishment. Chitosan, as a natural biopolymer, is currently widely used in plant tissue culture. The present study was conducted to overcome this problem in walnut under in vitro culture conditions. The experiment was performed in a completely randomized design with six replications for each treatment. The treatments included the control (Murashige and Skoog culture medium), thidiazuron at a concentration of 0.4 mg L⁻¹, polyvinylpyrrolidone at a concentration of 0.3 g L⁻¹, chitosan at a concentration of 0.1 mg L⁻¹, a combination of TDZ, PVP, and chitosan (at the concentrations used in the previous treatments), and finally chitosan nanocomposite optimized with PVP and TDZ at two concentrations of 0.1 and 0.05 mg L⁻¹. The chitosan nanocomposite at both concentrations significantly increased meristem survival (p < 0.05). Regeneration rate, as an indicator of vegetative growth, was 10% higher in the chitosan nanocomposite treatment than in the other treatments. In addition, shoot proliferation was significantly increased (p < 0.05) in this treatment compared with the other treatments.
